Zunächst studierte Aykroyd Kriminologie und Soziologie. 1972 trat Dan Aykroyd erstmals in einer Serie von 15-minütigen Comedyshows bei einem TV-Sender auf. Bereits hier schrieb er seine Sketche selbst. Dan Aykroyd war Mitbegründer der bekannten Fernseh-Reihe Saturday Night Live bei NBC. Viele Darsteller haben hier ihre Karriere begonnen. Zusammen mit John Belushi gründete Dan Aykroyd die Blues Brothers Band. Es folgten drei erfolgreiche LPs und eine Tournee. Dan Aykroyds größter Erfolg wurde dann auch der Film Blues Brothers. Mit Bill Murray drehte er Ghostbusters - Die Geisterjäger (1984) und Ghostbusters II (1989). 1990 kam die erste Oscar Nominierung als bester Nebendarsteller in Miss Daisy und ihr Chauffeur, der als bester Film ausgezeichnet wurde. Aykroyd spielte zuletzt eine Nebenrolle in James Belushis Sitcom Immer wieder Jim (bislang 4 Folgen).
